For what its worth _Burke's American Families_ states that:

John Saltonstall ... was brother of Gilbert Saltonstall (1495-1545) (ancestor
of Sir Richard Saltonstall, Lord Mayor of London 1597-98). John was b 1492
at High Saltonstall, d. 1559, leaving a son,

Gilbert Saltonstall, of Rookes Hall, b. 1525; d. 1598, having had, by
Margaret Harrison, his wife, two sons and two daus. of whom the elder son,

Samuel Saltonstall, of Rookes Hall and Huntwicke Grange, Hipperholm,
Yorkshire, b. 1562; m. 1st, Anne, dau. of John Ramsden, of Longley, and had issue, two
sons, 1. Richard, and 2, Gilbert. He m2 Elizabeth Ogden and d 8 Jan 1613
having had further issue.

This last Richard is said to be the founder of this branch in America. He
was b 1586; m1 1609 Grace d of Robert Kaye, m2 Elizabeth West, and died 1661.
More information given in this work and also further information in PA2 where it states that this Richard was _First Associate, Massachusetts Bay Comapny_

Thus the First Associate, Massachusetts Bay Comapny Saltonstalls would seem to be a different branch to the Saltonstall Alderman (and Lord mayor) of London
